چکیده

Gale, Glen R. (Veterans Administration Hospital, Durham, N.C.) and Helen H. McLain. Effect of ethambutol on cytology of Mycobacterium smegmatis. J. Bacteriol. 86:749-756. 1963.-Electron microscopy showed the effects of the antimycobacterial drug, ethambutol [d-2,2'-(ethylenediimino)-di-1-butanol], on the cytology of Mycobacterium smegmatis. After 10 hr of exposure to the drug, cells no longer contained material which, in control cells, was closely associated with cellular division, possibly genetic substance. No marked changes were observed in other cellular structures. It was concluded that the drug may block one or more steps in the synthesis of pentose nucleic acid or deoxypentose nucleic acid, ultimately causing cessation of cellular division and loss of viability. These cytological findings were compatible with results of other investigators on the mode of action of this drug.

Protection of Mycobacterium smegmatis from Ethambutol and Streptomycin Inhibition by MgSO(4) and Polyamines.

Polyamines protected Mycobacterium smegmatis from ethambutol inhibition but not from streptomycin inhibition, whereas MgSO(4) neutralized growth inhibition by both drugs.

Nonspecific ionic inhibition of ethambutol binding by Mycobacterium smegmatis.

Magnesium sulfate and spermidine were tested for their effects on binding of (14)C-ethambutol by Mycobacterium smegmatis. Concentrations were used that protected the organism from ethambutol inhibition. Sodium salts were examined as possible ethambutol antagonists to test the previously reported specificity of the divalent cation salt effect. Consistent with growth-protection experiments, 20 mM...

Ethambutol inhibition of glucose metabolism in mycobacteria: a possible target of the drug.

The addition of D-arabinose, D-galactose, D-glucosamine, or D-mannose to the growth medium of Mycobacterium smegmatis suppressed the inhibitory effects of ethambutol both on acetate labeling of cell wall-linked mycolic acids and on the increase in the delipidated cell dry weight. The addition of D-glucose or D-fructose had no effect. It is proposed that ethambutol inhibits an early step of gluc...

Development of a quantitative assay for mycobacterial endogenous arabinase and ensuing studies of arabinase levels and arabinan metabolism in Mycobacterium smegmatis.

Treatment of either Mycobacterium tuberculosis or M. smegmatis with ethambutol results both in inhibition of arabinan synthesis and in copious loss of previously formed arabinan from the cell wall. The loss of arabinan has been shown to be due to the action of an endogenous arabinase. To better understand this phenomenon, a quantitative assay for endogenous arabinase was developed. Using the as...
